摘要
目的探讨单胺氧化酶A(monoamine oxidaseA,MAO—A)基因rs5906893、rs1137070位点基因多态性与暴力行为的关系。方法采用聚合酶链式反应-限制性片段长度多态性(PCR—RFLP)的方法,检测92名某监狱暴力犯罪人员(暴力组)和102名正常对照者(对照组)的MAOA基因家族中的rs5906893位点和rs1137070位点的基因型。应用SPSS13.0分析基因多态性与暴力行为的关联性,应用遗传统计学软件进行2个SNPs的双位点联合分析。结果MAOA基因rs5906893位点暴力组、对照组CC基因型频率分别为63.0%、63.4%,TT基因型频率分别为37.0%、36.6%,频数分布无统计学意义(X^2=0.002,P=0.963,p〉0.05);rs1137070位点暴力组、对照组CC基因型频率分别为40.2%、36.6%,TT基因型频率分别为59.8%、63.4%,频数分布无统计学意义(x^2=0.262,P=0.609,P〉0.05);双位点联合分析结果显示,MAO—A基因的rs5906893位点和rs1137070位点T—T等位基因在暴力组和对照组中的分布差异有显著性(F=9.351,P=0.002)。结论MAO—A基因rs5906893位点、rs1137070位点单个基因多态性可能与暴力行为无关。但双位点分析显示两位点的T—T单倍型可能与暴力行为有关联。
Objective To investigate the association between monoamine oxidasc A (MAOA) gene rs5906893, rs1137070 polymorphism and violent behavior. Methods The methods of PCR-restriction fragment length polymorphism (RFLP)was conduetcd to detect rs5906893 and rs1137070 single nucleotide polymorphism loci of MAOA gene of in a 92 prison staff ( case group) and 101 normal controls ( control group). The association between the polymorphisms and violent behavior was analyzed with SPSS13.0. The UNPHASED program was applied to perform the combined effect analysis of the paired SNPs. Results MAOA gone rs5906893 of CC genotype frequency in the case group and the control group were 63.0% and 63.4% ,TT genotype frequency were 37.0% and 36.6% ,no significant differences were found between the two groups (P 〉 0.05). MAOA gene rs1137070 of CC genotypc frequency were 40.2% and 36.6% ,TT genotype frequency were 59.8% and 63.4% ,and no significant differences were found between the case and the control (P 〉 0.05 ). The results of the combined effect analysis showed that rs5906893-rs1137070 was associated with violent behavior(P= 0. 002). Conclusion MAOA gene rs5906893 ,rs1137070 polymorphism may be not associated with violent behavior. But the combined effect analysis showed that the haplotype of the paired SNPs was associated with violent behavior.
