摘要
目的探讨bFGF促愈合的作用机制。方法在新西兰大白兔背部制作创伤模型,外用bFGF(1000U/ml),进行肉眼观察、病理学检查、免疫组化分析和电镜观察,并用0.02mol/L的PBS做对照。结果肉限观察发现bFGF组表皮、肌肉的愈合率明显比阴性对照组高;病理学检查表明bFGF能促进肉芽组织中的成纤维细胞、DNA,毛细血管、胶原纤维的增长;免疫组化分析结果表明bFGF能显著地增加肌肉组织中肌动蛋白的数量,有利于伤口的收缩,电镜观察发现bFGF组肉芽组织中细胞功能旺盛。结论bFGF具有显著地促进创伤愈合的作用,其作用机制可能与以下几个方面有关:(1)促进创面巨噬细胞的增生、活化,吞噬功能增强;(2)促进表皮增生、伤口收缩;(3)促进肉芽组织生长。
Objective To study the mechanism of bFGF promoting wound healing. Methods 1000 U/ml bFGF was given to the wounded back of the New Zealand Big White Rabbits. The granulation tissues in the wound were observed by electron microscope and immunohistochemistry. Results bFGF could obviously promote the proliferation and activation of phagocytes in the early granulation tissues, phagocytose and clear the necrotic tissues,and elevate the growth of capillaries and fibroblasts and the synthesis of DNA in the granulation tissues. Immunohistochemically, bFGF could promote the proliferation of myofibroblasts and significantly increase the contents of actin in the experimental group. Microscopically,in experimental group,the granulation tissues contained a large number of myofibroblasts and various cellular biological functions were very active. Conclusion bFGF could promote the growth of the granulation tissues and the constriction of the wound and had a strong promotion on the would healing.
