摘要
大泷六线鱼主要分布在我国的黄海和渤海水域,是北部海区重要的经济鱼类之一。本文采用免疫组织化学方法和激素放射免疫测定技术,研究了血浆睾酮(T)和雌二醇(E2)含量、雄激素受体(AR)和雌激素受体(ER)在性腺组织中的定位,以及它们与性腺发育周期的关系。结果表明:雄鱼血浆T和E2含量表现明显的季节变动,在繁殖期(11月至翌年1月)内显著上升,T和E2水平分别达到21.26 ng/dL和13.36 pg/mL,在非生殖季节(2~8月)显著下降。ER在精巢间质细胞中表现强阳性反应,AR只在间质细胞中表现中等强度的反应。在恢复期发育的精巢(Ⅲ期),芳香化酶表现出强的免疫阳性反应,特别是在精原细胞和精母细胞中,在成熟精巢中阳性反应减弱,在精子细胞和精子中表现阴性反应。上述研究结果说明,睾酮、雌二醇及芳香化酶参与了大泷六线鱼精巢发育和繁殖内分泌功能调节。
Hexagrammos otakii is distributed mainly in the Bohai and Yellow Seas in China, and is a kind of important commercial Scorpaeniformes fish. In this paper, the main sexual steroid hormone in plasma, steroid hormone receptors and aromatase were studied by the methods of immunohistochemical techniques and radioimmunoassay methods. In the male, the level of plasma steroid hormone displayed seasonal variation, the main value of steroid hormone went up to peak in reproductive period from November to late January, the plasma testosterone(T) level increased significantly to 21.26 ng/dl, and 17β-estradiol(E2) level increased significantly to 13.36 pg/mL, and the plasma steroid hormone began to drop significantly from later February to August. In the male, the immunopositive response of estrogen receptor(ER) was located in the non-germinal cells, both Leydig and cell in the testis, showed strong immunopositive reaction. Immunopositive reaction of androgen receptor(AR) was located only in the Leydig cell, and showed medium positive reaction. The recover stage of testis development showed strong immunopositive reaction to aromatase antibodies, especially in spermatogonia and spermatocyte. The immunopositive reaction in spermatocyte to aromatase antibodies became weak gradually during the development and maturation of the testis, the spermatid and sperm showed negative immunopositive reaction. These results provide a new morphological proof that sex steroid hormones and aromatase participate in the regulation of gonadal development and function of reproductive endocrine in male Hexagrammos otakii.
