通塞颗粒对大鼠COPD急性加重期模型炎症细胞和肺功能的影响 被引量:4

Influence of Tongsai Granules on inflammatory cells and lung function in rat model of COPD in acute exacerbation stage

摘要 目的探讨通塞颗粒对大鼠慢性阻塞性肺疾病(COPD)急性加重期模型炎症细胞的影响。方法Wistar大鼠分为7组:对照组、COPD稳定期模型组、COPD急性加重期模型组、通塞颗粒高剂量组、通塞颗粒低剂量组、氨茶碱组、葶贝胶囊组,各10只。以反复呼吸道细菌感染的方法建立COPD急性加重期模型。对照组和模型组给以灌胃生理盐水,每日2次。通塞颗粒高、低剂量组、氨茶碱组、葶贝胶囊组分别给以中药通塞颗粒5g/(kg.d)、中药通塞颗粒2.5g/(kg.d)、氨茶碱2.3mg/(kg.d)、中药葶贝胶囊0.476g/(kg.d)灌胃,每日2次,给药时间为1周。观察通塞颗粒对外周血、支气管肺泡灌洗液中炎症细胞、肺功能的影响。结果外周血白细胞、中性粒细胞计数显示急性加重期模型组高于空白组和稳定期模型组(P<0.01,P<0.05);高剂量组低于氨茶碱组(P<0.05),外周血中性粒细胞计数显示高剂量组低于葶贝胶囊组(P<0.05)。高、低剂量组明显低于急性加重期模型组(P<0.01,P<0.05);白细胞和中性粒细胞计数显示稳定期模型组支气管肺泡灌洗液中高于空白对照组(P<0.05,P<0.01)、低于急性加重期模型组(P<0.01);高剂量组、低剂量组明显低于急性加重期模型组(P<0.05,P<0.01)和葶贝胶囊组(P<0.01,P<0.05);高剂量组支气管肺泡灌洗液中中性粒细胞明显低于氨茶碱组(P<0.01)。造模各组大鼠潮气量(Vt)、分钟通气量(MVV)、FEV0.3/FVC和呼气峰流速(PEF)明显低于空白对照组(P<0.01);急性加重期模型组、通塞颗粒低剂量组、葶贝胶囊组、氨茶碱组Vt、MVV、FEV0.3/FVC%和PEF均明显低于稳定期模型组(P<0.01)。通塞颗粒高剂量组、低剂量组、氨茶碱组Vt明显高于急性加重期模型组(P<0.05),通塞颗粒高剂量组MVV、FEV0.3/FVC和PEF均明显高于急性加重期模型组(P<0.05),吸气阻力和呼气阻力均低于急性加重期模型组(P<0.05);通塞颗粒高剂量组Vt、MVV明显高于葶贝胶囊组 Objective To explore the influence of Tongsai Granules on inflammatory cells and lung function in rat model of chronic obstructive pulmonary disease (COPD) in acute exacerbation stage. Method Wistar rats (n = 70) were randomly divided into 7 groups, including the control group (group 1 ), model group of COPD in stable stage ( group 2), model group of COPD in acute exacerbation stage (group 3 ), high-dose Tongsai Granules group (group 4 ), low-dose Tongsai Granules group (group 5 ), aminophylline group ( group 6) and Tingbei Capsules group ( group 7, each n = 10). The model of COPD in acute exacerbation stage was established by repeatedly bacterial infection of respiratory tract. Group 1,2 and 3 were given normal saline solution intragastrically twice a day, and group 4, 5, 6 and 7 were given intragastrically Tongsai Granules (5 g/kg/d and 2. 5 g/kg/d), aminophylline (2.3 mg/kg/d), and Tingbei Capsules (0. 476 g/kg/d) respectively twice a day. The medicine administration time was one week. The influences of Tongsai Granules on inflammatory cells in peripheral blood (PB) and bronchoalveolar lavage fluid (BALF), and lung function were observed. Result In group 3 the counts of white blood cells (WBC) and neutrophil (N) in PB were higher than those in group 1 and 2 (P 〈0.01, P 〈 0.05), and those were higher in group 4 than those in group 6 (P 〈 0.05 ). The count of N in PB was higher in group 4 than that in group 7 (P 〈 0.05). In group 4 the counts of WBC and N in PB were lower than those in group 6 (P 〈 0.05 ). In group 2 the counts of WBC and N in BALF were higher than those in group 1 (P 〈0.05, P 〈0.01 ), and lower than those in group 3 (P 〈0.01 ). In group 4 and 5 the counts of WBC and N in BALF were significantly lower than those in group 3 (P 〈0.05, P 〈0.01 ) and group 7 (P 〈0.05, P 〈0.01 ).
