藤黄酸诱导SK-MES-1肺鳞癌细胞凋亡

摘要 目的研究藤黄酸(GA)对人肺鳞癌细胞株(SK-MES-1)的生长抑制、诱导凋亡作用及分子机制。方法采用改良四甲基偶氮唑盐比色法(MTT)和流式细胞术检测GA对SK-MES-1的生长抑制及细胞凋亡作用,Western-blot法检测不同浓度GA作用SK-MES-1后细胞凋亡蛋白酶Caspase9、10及p53蛋白表达。结果GA对SK-MES-1有明显的生长抑制和促凋亡作用;GA呈浓度依赖性上调Caspase9、10及p53蛋白的表达。结论GA具有显著的促SK-MES-1肺鳞癌细胞凋亡作用,其分子机制可能与上调Caspase9、10及p53蛋白的表达有关。 Objective To investigate the inhibitory effects of gambogic acid (GA) on the growth and apoptosis of SK-MES-1 lung squamous cancer cells. Methods Modified MTT assay and flow cytometry were used to detect the growth and apoptosis of SK-MES-1 cells induced by GA. Western blotting was used to detect the expression levels of Caspase 9,10 and p53 protein. Results The growth of SK-MES-1 cells was significantly inhibited and its apoptosis increased by GA. The expressions of Caspaseg, 10 and p53 were up-regulated concentration-dependently by GA. Conclusion GA can promote the apoptosis of SK-MES-1 lung squamous cancer cells, which may be related to the up-regulation of the expressions of Caspase 9,10 and p53.
