摘要
目的探讨一种新的凋亡抑止蛋白Livin在人脑星形细胞瘤中的表达及其对瘤细胞增殖和凋亡的影响。方法分别采用免疫组织化学和原位细胞凋亡检测法检测人脑胶质瘤细胞中Livin、PCNA蛋白的表达和凋亡细胞,结合胶质瘤病理分级分析其相关关系。结果随着人脑胶质瘤WH0病理分级的增高,Livin、PCNA的表达强度明显升高(P〈0.05),细胞凋亡率也明显升高(P〈0.05),其中高度恶性组(WHO分级Ⅲ、Ⅳ级)均显著高于低度恶性组(WHO分级I、Ⅱ级)(P〈0.05);随着Livin表达强度的增高,PCNA表达强度及细胞凋亡率均增高,但PCNA的表达比细胞的凋亡有更明显的增高趋势。结论人脑星形细胞瘤中Livin、PCNA的表达强度和细胞凋亡率异常升高;星形细胞瘤的发生和演变可能与Livin、PCNA及细胞凋亡的综合作用密切相关。
Objective To investigate the expression of Livin and its relationship with pathological grades, proliferation and apoptosis of glioma cells. Methods The expression of Livin, PCNA and apoptotic cells were detected by immunohistochemistry and TUNEL assay in 55 samples of human brain gliomas and 12 samples of normal brain tissue. Results Livin, PCNA and apoptotic cells were not expressed in normal brain tissue. The expression of Livin was increased in human brain glioma with the advancing of pathological grades. There are significant differences in the expression of Livin and PCNA, and apoptotic cells between high grade (grade Ⅲ and Ⅳ) and low grade (grade Ⅰ and Ⅱ) tumors(P 〈0.05). Conclusion There are increased Livin, PCNA expression in human brain gliomas, which may be associated with carcinogenesis and progression and cell apoptosis.
