联合补充乙酰左旋肉碱和α-硫辛酸对耐力训练大鼠骨骼肌线粒体通透性转运孔开放的影响 被引量:2

摘要 观察联合补充乙酰左旋肉碱(acetyl-L-catnitine,ALCAR)和α-硫辛酸(α-lipoic acid,LA)对耐力训练大鼠骨骼肌线粒体通透性转运孔(MPTP)开放的影响。方法:24只SD大鼠随机分成对照组、训练组和训练+给药组。训练组进行无负重游泳训练,每周6天,每天时间从45min逐渐增加到90min。补充方案为在饮用水中加入0.5%的ALCAR,pH值调至5.2,自由饮用;将食用饲料碾碎并加入0.2%的LA喂食。6周后,利用H2O2作为MPTP诱导剂,采用紫外分光光度仪检测骨骼肌线粒体PTP开放;采用荧光光度仪,通过测试介质Rh123的荧光值检测线粒体膜电位;采用双光束紫外分光光度仪,通过测试Ca2+指示剂AⅢ吸光值检测Ca2+转运。结果:(1)训练组PTP开放检测吸光值最高,其次是对照组,训练+给药组最低,但不同剂量H2O2诱导下,三组吸光值无显著性差异。(2)训练+给药组线粒体膜电位在诱导后与对照组和训练组比较均有显著性差异(P<0.05);同组诱导前后比较时,三组5μl诱导后和10μl诱导后与诱导前比较均有显著性差异(P<0.05,P<0.01),且对照组和训练组10μl诱导后与5μl诱导后相比有显著性差异(P<0.05),训练+给药组无显著性差异;(3)在线粒体Ca2+转运方面,三组5μl诱导后和10μl诱导后与诱导前比较均有显著性差异(P<0.05,P<0.01),且对照组和训练组10μl诱导后与5μl诱导后相比有显著性差异(P<0.05),训练+给药组无显著性差异。结果提示:联合补充ALCAR和LA可能会抑制H2O2诱导的MPTP开放,稳定线粒体内外的钙离子平衡。
