摘要
采用水酶法技术从菜籽中提取油和水解蛋白,对工艺中蛋白酶酶解参数(温度、酶量、酶作用时间、pH)应用因子设计响应面分析法进行优化。得到清油得率和水解蛋白得率与酶解参数之间关系的两个模型方程和酶解最佳参数条件:酶用量2.4%,酶解温度55.5℃,时间4.5 h,pH 8.5。在此条件下,清油得率和水解蛋白得率分别为90.12%和87.39%。水解蛋白相对分子质量700 ua以下的短肽占88.7%。
An aqueous enzymatic extraction technology was adopted to extract oil and protein hydrolysates from rapeseeds.The protease hydrolysis conditions (viz., temperature, enzyme to substrate level, time and pH) for recovery oil and protein hydrolysates wore optimized by response surface methodology using a factorial design. Two mathematical models of free oil and pro- tein hydrolysate yields were obtained with regard to the effect of temperature, enzyme to substmte level, time and pH. An enzyme to substrate level of 2.4%, temperature 55.5 ℃, ahydrolysis time 4.5 h and pH 8.5 were found to be the optimum conditions to obtain relatively high free oil yield and protein hydrolysate yield of 90.12% and 87.39% respectively. Results show 88.7% peptides in the protein hydrolysates are oligopeptides with molecular weight lower than 700 u.
