亚硒酸钠对人早幼粒白血病细胞的RB蛋白磷酸化的影响 被引量:1

摘要 以人早幼粒白血病细胞(HL-60)为实验对象,利用Western-blotting方法和流式细胞仪分析,研究亚硒酸钠对HL-60细胞的RB蛋白产物磷酸化的影响和探讨其抗癌机制。Western-blotting结果显示:与对照组相比,在第四天5.8μmol/LNa2SeO3可使磷酸化Rll蛋白减少49%,去磷酸化RB蛋白仅轻度增加,但在加硒组中去磷酸化RB蛋白占RB蛋白总量的71.1%。与此同时,细胞周期的结果表明:亚硒酸钠对HL-60细胞的G2+M和S期有一定的阻滞。该实验在蛋白分子水平上研究了亚硒酸钠抑制HL-60细胞增殖的作用机制。 Human promyelocytic leukemia cells (HL-60) was used to study the effect of sodiumselenite on phosphorylation of retinoblastoma (RB) protein and its anticancer mecha-nism. Method of western--blotting and flow cytometric analysis were undertaken. Theresult of western-blotting showed that 4 days after treatment of 5. 8μmol/L. Na2SeO3,the amounts of the phosphorylated RB protein decreased some 49%that of controlgroup,while the hypophosphorylated RB protein increased a little. However, in this selenium group the phosphorylated RB protein was 71.1 % in total RB protein amounts.In the same time, the result of cell cycle revealed that sodium selenite could slightly arrest HL-60 cells at G2+M and S phase. The experiments studied the mechanism of sodium selenite inhibited HL-60 cells proliferation at protein molecules level.
