摘要
为探索硒的抗白血病机理,观察了硒代二半胱氨酸对白血病U937和K562细胞cAMP、cGMP及细胞周期的影响。结果表明,用3.0μmol/L硒代二半胱氨酸作用3天后,白血病细胞生长和增殖被抑制,其细胞内cAMP含量增加,cGMP含量减少,cAMP/cGMP比值在U937和K562细胞分别从0.24和2.08提高到1.53和4.77,S+G2/M期细胞百分比分别减少3.8%和12.3%。
n this article,the effects of dicysteine selenium were
observed on the level of cAMP,cGMP and cell-cycle in leukemia cell lines(U937 and K562 ).After
incubation for 3 days with dicysteine selenium at aconcentration of 3.0 μmol/LIC50, the level of
cGMP decreased significantly(P<0.05),and the level ofcAMP increased markedly(P<0.01),when
compared with the controls. Meanwhile the ratio of the num-ber of cells in S + G2/M phases
decreased significantly after the addition of dicysteine selenium. It wasdemonstrated that
dicysteine selenium can inhibit DNA systhesis and division of U937 and K562 leukemiacell
lines with a higher ratio of cAMP/cGMP.
