反义核酸和维甲酸对早幼粒白血病细胞NB4的诱导分化

摘要 目的:探讨反义核酸和维甲酸对早幼粒白血病细胞株NB_4细胞生长分化的影响.方法:采用细胞生长曲线、形态学、细胞表面标志及硝基四氮唑蓝(NBT)还原试验判定NB_4细胞的生长、分化及功能.结果:PML-RARα基因的反义核酸和全反式维甲酸能够诱导NB_4细胞分化,提高NBT还原率,降低细胞表面的CD33抗原,升高CD11b抗原.反义核酸更有利于CD33抗原的降低,而全反式维甲酸更有利于CD11b抗原的升高.结论:PML-RARα基因的反义核酸和全反式维甲酸能诱导NB_4细胞分化,但它们的作用机制可能不同. To study the effects of antisenseoligodeoxynucleotides (ASODN) specialized to PML-RARα gene or all-trans retinoic acid(ATRA) on proliferation and differentiation in acute promyelocytic leukemia cell line NB4 cells. Methods: Proliferation curve, morphology, CD antigen of membrane and NBT test were used to determine the proliferation and differentiation of NB4 cells. Results: ASODN or ATRA could differentiate acute promyelocytic leukemia cell NB4, improve positive rate of NBT, up-regulate GD1 1b antigen and down-modulate CD33 antigen. In contrast, ASODN had more down-modulation of CD33 antigen and less up-regu-lation CD11b antigen than ATRA. Conclusion; ASODN spe-cialized to PML-RARa gene or ATRA can both differentiate acute promyelocytic leukemia cell NB4, but maybe have different mechanisms.
