摘要
采用PCR-SSCP技术测定了4个黄羽肉鸡品种IGF-1基因2个SNP位点,并分析了它们对不同周龄体重及产蛋性状的遗传效应。结果显示,2个SNP位点分别发生了T→A、G→A的单碱基突变,各有3种基因型,京海黄鸡P1位点偏离Hardy-Weinberg平衡状态(P<0.01)。最小二乘方差分析表明,京海黄鸡、江村黄鸡AA型个体具有较高的300日龄产蛋量,与AB、BB型间差异显著(P<0.05),江村黄鸡AA型具有较高的12周龄和300日龄体重,石岐杂鸡AA型300日龄体重显著高于BB型(P<0.05),表明A为产蛋及体重增效基因;不同黄鸡品种均表现出CC型个体具有较高的300日龄体重和产蛋数(P>0.05)。
Two SNP loci of IGF-1 gene were detected by PCR-SSCP method in Jinghai Yellow Chicken,Jiangcun Yellow hens,Shiqiza hens and Liyang hens,and their genetic effects on production performances were also analyzed. The results showed the mutations of T→A ,G→A in P1 ,P2 loci were identified respectively,and there were three genotypes in each locus,three genotypes in Pt locus in Jinghai Yellow chicken were deviated the Hardy-Weinberg Equilibrium (P〈0.05). The Least Square Means analysis showed,AA individuals in Jinghai Yellow Chicken,Jiangcun Yellow hens had more eggs than AB and BB ones in 300 days (P〈0.05),and AA Jiangcun Yellow individuals had higher 12-week and 300-day weight,and AA Shiqiza individuals had higher 300-day weight than BB ones (P〈0.05),and A was an egg-number and body weight enhancing gene, B was a leaky one. CC individuals in 4 breeds were all have higher 300-day weight and egg number (P〉0.05).
