摘要
目的:探讨一次性力竭运动后大鼠心房肌蛋白质组的表达特征,初步筛选对力竭运动应激有意义的心房肌蛋白质。方法:10只SD雄性大鼠随机分为对照组和运动组。采用三级递增运动负荷跑台训练建立动物力竭运动模型。提取心房肌组织全蛋白进行双向电泳分离。结果:经图像分析,对照组平均检测到382±24个蛋白质点,运动组平均检测到352±17个点。与对照组相比,运动组共有47个蛋白质点发生了变化。12个点在运动后消失,运动后表达量上调3倍以上的点有25个,下调超过2/3的有10个,其中上调量在5倍以上的有14个,下调超过4/5以上的有6个。对运动后表达差异大的4个蛋白质点进行质谱鉴定,鉴定出心肌型-α肌球蛋白重链、乌头酸水合酶、异戊酰辅酶A脱氢酶和甘露糖结合蛋白C前体4种蛋白质。结果表明,一次性力竭运动后,大鼠心房肌的蛋白质组发生了明显改变。
Objective To study the expressive feature of atrial muscle proteome after a single bout of exhaustive exercise and primarily screen out some meaningful responsive proteins to exercise stress. Methods Ten rats were randomly divided into control group(n=5)and exercise group(n=5). After 3-day adaptive treadmill training,the rats in exercise group were running on treadmill with Bedford 3-grade incremental protocol until exhaustion,and then were killed for extracting the total protein of atrial muscle using 2D electrophoresis (2-DE) in combination with immobilized pH gradients separation. The disappeared protein spots and spots with differential expression volume more than 5 folds were selected for mass spectra(MS)identification. Results and Conclusion In electrophoresis map, the average number of detected protein spots in control group and exercise group was 382 ±24 and 352 ±17, respectively. After exercise,47 protein spots changed:12 disappeared;14 increased more than 5 folds;10 decreased...更多 more than 2/3. Four protein spots were selected for MS identification,and 4 types of proteins including. mannose-binding protein C precursor,isovaleryl-CoA dehydrogenase and aconitate hydratase,were identified. The results indicated the marked changes in myocardium proteome after exhaustive exercise.
