β--七叶皂甙钠对实验性脑出血后AQP4表达及脑水肿的影响 被引量:4

Effect of β--sodium aescinate on aquaporin--4 expressions and cerebral edema in experimental rats with intracerebral hemorrhage

摘要 目的研究β--七叶皂甙钠对大鼠脑出血后水通道蛋白4(AQP4)表达及脑水肿的影响,探讨β--七叶皂甙钠治疗出血性脑水肿的作用机制。方法在大鼠苍白球注射胶原酶制作脑出血模型,采用干湿重法观察脑水肿的变化;应用免疫组织化学方法检测脑组织AQP4表达。结果脑出血模型大鼠病灶侧脑含水量、血肿周围AQP4的表达水平均明显高于假手术组(均P〈0.01);各个时间点β--七叶皂甙钠治疗组病灶侧脑含水量以及血肿周围AQP4的表达水平均明显低于对照组(均P〈0.05);AQP4表达水平与脑含水量呈正相关(r=0.976,P〈0.01)。结论β--七叶皂甙钠能明显减轻脑出血后脑水肿及AQP4的表达,β--七叶皂甙钠可能抑制AQP4的表达而减轻脑水肿。 Objective To investigate the effect ofβ--sodium aescinate on aquaporin--4 expression and cerebral edema of rats with intracerebral hemorrhage and to explore the mechanisms of β--sodium aeseinate in treatment of hemorrhagic brain edema. Methods The model of intraeerebral hemorrhage (ICH) was established by infusing collagenase into globus pallidus of the rats. Dynamic change of brain edema was observed using wet--dry weighing method. Immunohistochemieal staining was used to determine the expression of cerebral AQP4. Results Brain water content (BWC) of injured side and the AQP4 expression levels of perihematom tissue in ICH rats were significantly higher than that of the sham operation group (P 〈 0.01). However, the BWC and AQP4 expression levels were remarkably reduced in theβ--sodium aescinate group compared to the control group at the indicated time points (P 〈 0.05). The expression levels of AQP4 positively correlated with BWC (r = 0. 978, P 〈0.01). Conclusions AQP4 expression is greatly elevated in the rats with cerebral edema, and that may contribute significantly to the development of cerebral edema after ICH. β--sodium aescinate can relieve cerebral edema after ICH via restraining AQP4 expressions.
