摘要
目的探讨大鼠蛛网膜下腔出血后脑水肿组织中水通道蛋白4(aquaproin 4,AQP4)的表达变化。方法采用自体血注入枕大池法建立蛛网膜下腔出血动物模型,原位杂交和免疫组化法的方法分别检测蛛网膜下腔出血后AQP4 mRNA和蛋白质表达变化,并用干湿重法测定脑含水量变化。结果蛛网膜下腔出血后8h,脑含水量和AQP4 mRNA及蛋白表达开始上升,2d达到高峰,1周后降低到正常水平;AQP4 mRNA及蛋白的表达和脑含水量呈显著正相关(r=0.9236~0.9509,P<0.01)。结论AQP4与蛛网膜下腔出血性脑水肿的形成密切相关。
Objective To investigate the expression of aquaproin-4 (AQP4) in rat brain after subarachnoid hemorrhage (SAH). Methods The subarachnoid hemorrhage model was induced by injection of autologous blood into the rat's cisterna magma. In situ hybridization and immunohistochemistry was used to determine the change of expression of AQP4 mRNA and AQP4 protein during different periods after SAH. The changes of brain water content (BWC) were measured by the wet and dry weight methods. Results The BWC, expression of AQP4 mRNA and AQP4 protein in rat cortex after SAH were increased at the 8th hour and reached peak on the 2nd day and went to the normal level on the 7th day . The expression of AQP4 mRNA and protein was positively correlated with BWC ( r = 0. 9236 - 0. 9509, P 〈 0.01 ). Conclusions AQP4 has a relevant relationship with the formation of brain edema after SAH.
