摘要
目的探讨非小细胞肺癌(NSCLC)组织中PTEN、VEGF、TSP-1的表达及与肿瘤血管生成的关系。方法采用免疫组化(SP法)分别检测70例NSCLC组织和12例正常肺组织PTEN蛋白情况,以及不同PTEN表达组的肺癌组织中血管内皮生长因子(vascular endothelial growth facfor,VEGF)、血小板反应素-l(thromhospondin-1,TSP-1)的表达及微血管密度(MVD)值的变化。结果NSCLC组织中PTEN的阳性表达率为54.29%,低于其在正常肺组织中的阳性表达率100%(P<0.05);PTEN阳性组MVD值40.42±3.26低于PTEN阴性组的MVD值55.31±7.24(P<0.05)。NSCLC组织中PTEN与VEGF的表达强度成负相关(r=-0.458,P<0.05);PTEN与TSP-1的表达强度成正相关(r=0.836;P<0.01)。结论NSCLC组织中PTEN表达降低;PTEN阳性组MVD明显降低;PTEN通过抑制VEGF表达并上调TSP-1表达来抑制肿瘤血管生成,导致肺癌预后不良。
Objective To study the relationship between the expression of PTEN, VEGF and TSP - 1 and tumor angiogenesis in non - small cell lung carcinoma(NSCLC). Methods Immunohistochemical technique was used to de- tect the expression of PTEN, VEGF and TSP - 1 in 70cases of NSCLC and 12 cases of normal lung tissue, the microves- sel density(MVD) marked by CD34 was also detected. Results The rate of PTEN protein expression in experimental group (54.29%) is significantly higher than that of normal lung tissue( 100% ), there was a significant difference in them (P 〈0.001 ). In NSCLC,MVD of PTEN- positive group (40.42 ±3.26) was significantly lower than that of PTEN - negative group ( 55.31 ± 7.24) ( P 〈 0.05). PTEN and VEGF expression in NSCLC was negatively correlated ( r = - 0.458,P 〈 0. O1 ) ; PTEN and TSP - 1 expression in NSCLC was positively correlated ( r = 0. 836, P 〈 0.01 ). Conclusions In NSCLC, there was loss of PTEN expression. The lower expression of PTEN in lung carcinoma may lead to increasing of the MVD, and it isassociated with the bad prognosis of lung carcinoma. PTEN inhibited angiogene- sis in NSCLC by inhibiting the VEGF expression and increasing the expression of TSP - 1 at the same time.
