摘要
目的建立高效液相法测定血及尿中硝基安定的浓度。方法采用Agilent Eclipse XDB-C18色谱柱以甲醇-水(70∶30)为流动相,流速1.0mL·min-1,检测波长254nm。结果该法分离效果较好;硝基安定质控3种浓度(4.0,8.0,16.0μg.mL-1)日内和日间的RSD,血浆样品均小于7%,尿样均小于5%,血浆回收率为87.13%~92.15%,尿回收率为89.82%~92.75%,血浆及尿样样品线性范围为4.0~64.0μg.mL-1,相关系数在0.9972。血浆、尿样中硝基安定最低检出限为4.0μg.mL-1。结论此方法可对硝基安定进行中毒药物快速检测并定量。
Objective To establish an HPLC method to detect the concentration of nitrazepam from human plasma and urine. Methods Agilent Eclipse XDB-C18 was equipped, methanol and water (70 : 30) were used as the mobile phase. Flowrate was 1.0mL·min^-1, and the detection wavelength was 254nm. Results With three quality control concentrations of nitrazepam (4. 0, 8. 0 and 16.0μg ·mL^-1), RSD between inner day and daytime, samples from plasma were less than 7%, samples from urine were less than 5 %. The recovery rates were 87. 13%-92.15% for plasma and 89.82%-92. 75% for urine. The standard calibration curves of concentrations and peak area were liner in the concentration ranges of 4. 0-64.0μg ·mL^-1, and the correlation coefficient were 0. 9972 for plasma and urine respectively. Conclusions An accurate, convenient and easily-repeatedmethod for determination of troxerutinin plasma was established
