摘要
目的:研究递增负荷运动对大鼠心肌细胞凋亡调控基因Bcl-2和Bax的影响,探讨递增负荷运动对心肌细胞的作用。方法:将大鼠分为0Wk~5Wk运动组,每组又分为:安静组、运动后即刻组、运动后3h组和运动后24h组,负荷从20m/min递增到40m/min,采用反转录聚合酶链式反应(RT-PCR)技术,观察其基因的表达。结果:(1)各周运动后即刻,Bcl-2mRNA表达0Wk较1Wk、3Wk和5Wk显著升高,Bax表达5Wk较4Wk显著升高;(2)各周运动后3h,Bcl-2表达4Wk较2Wk极显著降低,Bax表达2Wk和5Wk分别较0Wk显著升高和降低;(3)各周运动后24h,Bcl-2表达各周都较0Wk有极显著性降低,Bax表达5Wk较0Wk和4Wk明显降低。结论:递增负荷运动后即刻,心肌表现出一种自我保护性的应激反应,运动后3h与24h,尤其是24h,心肌抗凋亡能力减弱。
Objective: This paper probed the effects of incremental exercise on apoptotic gene Bcl-2 and Bax in myocardiumin order to study the regulating mechanism of incremental exercise on apoptotic gene Bcl-2, Bax in myocardiumin. Method: The rats were divided into quiet group, immediate after exercise, 3h after exercise and 24h after exercise. The expression of genes Bcl-2, Bax and the Bcl-2/Bax ratio were recorded in sedentary state, immediately af- ter training and 3h and 24h after training were determined by RT-PCR. Results: (1)The expression of gene Bcl-2 determined immediately after training was higher in 1Wk and 5Wk as compared with that in 0Wk. The expression of gene Bax was higher in 5Wk than that in 4Wk. (2)At 3h after training the expression of gene Bcl-2 was significantly reduced in 4Wk as compared with that in 2Wk. The expression of gene Bax was lower in 0Wk as compared with that in 2Wk, also lower than that in 5Wk. (3)During 0Wk-SWk, at 24h after training the expression of gene Bcl-2 was the lowest in 0Wk. The expression of gene Bax was decreased in 5Wk as compared with that in 0Wk and 4Wk. Conclusion: The cardiomyocyte produced an active response to self-protect immediately after training. The anti-apoptosis ability of cardiomyocyte went down at 3h and 24h after training, especially at 24h.
