摘要 目的:通过建立兔耳增生性瘢痕模型,研究肿瘤坏死因子(tumor necrosis factor-alpha,TNF-α)在减轻增生性瘢痕形成中的作用。方法:建立兔耳增生性瘢痕模型,实验动物随机分为A、B、C、D、E五组,并设C组为对照组,A、B、D、E组瘢痕内分别注射不同浓度TNF-α溶液0.1ml(100ng/ml,500ng/ml,1000ng/ml,5000ng/ml),C组瘢痕内注射等量生理盐水。观察增生性瘢痕组织块的组织结构、成纤维细胞的密度以及caspase-3蛋白的表达情况。分析不同浓度TNF-α对增生性瘢痕形成的影响。结果:瘢痕增生块减轻程度随TNF-α浓度增加而增加,即TNF-α浓度越高,增生性瘢痕体积越小,成纤维细胞密度越低,caspase-3表达强度越强。结论:肿瘤坏死因子-α可明显抑制兔耳增生性瘢痕的形成及瘢痕组织增生。 Objective To investigate the inhabit efects of TNF-α (tumor necrosis factor -alpha) on the hypertrophic scar in rabbits ear model. Methods Fifteen Japanese white rabbits were used to establish the models of hypertrophic scar in ears.The rabbits were divided to five groups, A ,B,C,D,E.The rabbits in the experimental and control groups were received each 0.1 ml TNF-α (100ng/ml, 500ng/ml, 1000ng/ml, 5000ng/ml) and 0.1 ml normal saline. The effects of TNF- α on the hypertrophic scar were checked by HE staning and enzyme linked immunosorbent assay. According to observe the structure of hypertrophic scar , the number of fibroblasts changes and the expression of caspase-3 protein .To detect the effect of TNF-α on the hypertrophic scar with different consistency of rabbits ears. Results Compared with the control group, with the increase of TNF-α, the proliferation of fibroblast was obviously inhibited and the volume of hypertrophic scar were decreased in the TNF-α treated group.The expression of caspase-3 protein increased. Conclusion TNF-α can inhibit formation of hypertrophic scar in rabbit ears,
