响应面法及三波长技术优化蒽酮比色法测定豆类淀粉含量的研究 被引量:9

Optimizing Anthrone Colorimetric Method in Determination of Legume Starch by Response Surface Methodology & Three-Wavelength Technique

摘要 拟利用蒽酮比色法结合三波长技术测定豆类淀粉含量,在单因素实验的基础上,利用响应面法建立蒽酮比色法测定淀粉含量工艺的二次多项式数学模型,并在所建模型的基础上对测定条件进行优化。结果表明三波长技术优于单波长技术;在测定过程中以盐酸作为水解酸,避免了淀粉以外的其它碳水化合物的水解,使得测定结果更准确;单因素实验中各因素最佳条件为盐酸用量0.4 mL、水解水浴时间2 min、蒽酮用量0.5 mL、硫酸用量5 mL、显色时间3 min;建立的二次多项式数学模型具有很高的拟合度,据二次多项式数学模型优化出淀粉最佳测定条件为蒽酮用量0.5 mL、硫酸用量4.8 mL、显色时间18.8 min。优化建立的三波长蒽酮比色法测定淀粉含量的方法具有准确、简便、快捷、适宜于多个淀粉样品测定的特点。 The anthrone colorimetric method combined with three - wavelength technique was used to determine the content of starch in legume. Based on the results of single - factor experiments, the response surface methodology was used to establish a quadratic polynomial regression mathematics model on the determination of the starch content by the anthrone colorimetric methed, and then the model was applied to optimize the determining conditions. Results indicate three - wavelength technique is superior to one - wavelength technique. To avoid other carbohydrate besides starch being hydrolyzed, using hydrochloric acid as hydrolytic acid is proper to achieve accurate determination. The optimum factor conditions resulted from the single - factor experiments are hydrochloric acid dose 0.4 mL, hydrolytic time 2min, anthrone dose 0.5 mL, sulfuric acid dose 5 mL, and color display 3min; the optimum determining conditions resulted from the quadratic polynomial regression mathematics model are anthrone dose 0.5 mL, sulfuric acid dose 4.8 mL, and color diaplay 18.8min. The established method provides an exact, convenient and fast means to quantify the starch content in legume, meeting the requirement of multi - sample determination.
