连续流动法测定新鲜烟叶中的直链淀粉和支链淀粉 被引量:8

Determination of amylose and amylopectin of fresh tobacco by continuous flow method

摘要 烟叶中直链淀粉和支链淀粉的含量与烟叶的品质和产量有着重要的关系,检测新鲜烟叶中淀粉含量,对于掌握烟株生长和烟叶品质有重要意义.直链淀粉和支链淀粉是烟草中较难测定的成分.研究以高氯酸为提取剂,建立了一种快速、准确测定新鲜烟叶中直链淀粉和支链淀粉的分析方法.方法的操作步骤是,采用液氮研磨剪碎的鲜烟叶,用乙醇-饱和氯化钠溶液脱色,40%高氯酸溶液提取,直链淀粉采用在线氢氧化钠碱液分散二次进样显色,选择波长600 nm的滤光片,支链淀粉采用直接进样显色,选择波长570 nm的滤光片,连续流动法测定烟草中2种淀粉含量.直链淀粉和支链淀粉的检出限分别为0.49 mg/g和0.37 mg/g,平均回收率分别为111.97%和104.41%.方法对两种淀粉的相对标准偏差均在5%以内. Starch,including amylose and amylopectin,is very important to quality and yield of tobacco leaf.To detect the content of starch in fresh tobacco leaves is significance for master the growth and quality of tobacco leaves.However,amylose and amylopectin in tobacco are difficult to determine.An improved method using HClO_4 as extractant was established for fast and accurate determination of these two constitutes.The method included the following steps: grinding the cut fresh tobacco with liquid nitrogen,bleaching the sample uing alcohol-Na Cl saturated solution,extracting the starch from tobacco with 40% HClO_4,continuous flow analyzing at the wavelength of 600 nm and 570 nm for amylase and amylopectin,respectively.The amylose was colorized after sodium hydrate solution extraction while the amylopectin was colorized directly. The detection limits of amylose and amylopectin were 0.49 mg/g and 0. 37 mg/g,respectively,the average recovery for amylose and amylopectin were 111. 97% and 104.41% ,respectively,and the relative standard deviation of the two starches was both less than 5% .
