The role of GSK-3β in the metabolism of amyloid precursor protein

摘要 糖原合酶激酶-3β(GSK-3β)在细胞内参与了多个生理过程,其中其与阿尔茨海默病(AD)的关系值得关注,它不但是tau蛋白形成神经纤维缠结(NFT)的主要蛋白激酶,还是Aβ诱导tau蛋白过度磷酸化中的主要中介,其转录受到淀粉样蛋白质前体(APP)的影响。本文主要介绍了GSK-3β对APP代谢主要过程的参与及其作用,并简单介绍了一些GSK-3β抑制剂对AD的影响。 Alzheimer's disease (AD) is characterized by the intracellular accumulation of hyperphosphorylated tau into tangles and the extracellular accumulation of amyloidβ(Aβ) peptide into amyloid plaques. The glycogen synthase kinase-3beta (GSK-3β) participate in diverse cellular processes from diabetes and cancer to mood disorders and the neurodegeneration of AD. Not only can it be a major link in the signaling pathway from amyloid to taupatbology, but also it can exert an important effect on the cleavage of amyloid precursor protein (APP). On the other hand, APP and FE65, which is another protein influenced by the effect of GSK-3β on APP, may have a role as transcriptional regulators in nuclear. In this review we made a summary of the role of GSK-3β in Alzheimer's disease.
