摘要 阿尔茨海默病是一种神经退行性疾病,主要病理特征为细胞外间隙β淀粉样蛋白沉积所形成的老年斑;细胞内异常磷酸化tau蛋白聚集形成的神经纤维缠结;以及海马神经元丢失。铜离子与阿尔茨海默病的发生与发展密切相关。铜离子通过与淀粉样蛋白、淀粉样蛋白前体以及tau蛋白的相互作用,影响β淀粉样蛋白和tau蛋白的结构,介导细胞内信号通路,改变脑内的氧化平衡状态。本文综述了铜离子对阿尔茨海默病发生发展的神经活性作用,介绍了铜离子的稳态失调对阿尔茨海默病中β淀粉样蛋白和tau蛋白生理活性的影响。 Alzheimer's disease (AD) is a progressive neurodegenerative disease characterized by the extracellular accumulation of neurotoxic amyloid-β (Aβ), the intracellular accumulation of hyperphosphorylated tau and the loss of synapse. Copper ion plays a key role in AD development. The Aβ, amyloid precursor protein (APP) and tau protein are modulated by copper ion in the brain. Copper ion could affect the structure of Aβand tau protein, regulate cellular signal pathway and influence the oxidative balance in AD brain. This review explores the neuro-activity of copper ion in AD development and the effect of copper ion homeostasis to the bioactivity of Aβ and tau protein in AD brain.
