摘要 目的观察苦参碱(matrine)对体外大肠癌细胞凋亡发生及Bad蛋白表达的影响。方法大肠癌LOVO细胞株分为对照组和苦参碱不同浓度作用组,将终浓度分别为0.1、0.5、2.5g/L的苦参碱加入培养液中,观察大肠癌细胞形态学变化,测定各组苦参碱作用1、4、16h后的细胞活力(MTT法)、细胞凋亡率(流式细胞术)、Bad蛋白含量(免疫组化法和western blot法)。结果与对照组比较,苦参碱组出现细胞皱缩、脱壁、细胞漂浮及细胞破碎现象,细胞活力显著降低,细胞凋亡率增加,Bad蛋白表达增加,有明显的时间和浓度依赖关系。结论苦参碱能提高大肠癌细胞凋亡发生率和Bad蛋白的表达。 Objective To observe the effect of matrine on apoptosis and Bad expression of colorectal cancer cells in vitro. Methods Colorectal cancer cells were divided into the normal group and the matrine group in which matrine of different content (0. lg/L,0.5g/L,2.5g/L) were added for 1,4 and 16 hours. The morphological changes of cancer cells were observed. Cell vigor(MTT method), apoptosis rate(Flow eytometry), Bad expression(immunohistochemistry and western blot) in each group were assayed respectively. Results In the matrine group, shrinkage cells, floating cells and broken cells were found. Compared with the normal group, the cell vigor in the matrine group was significantly lower while apoptosis rate,Bad expression were significantly higher time-dependently and dose-dependently. Conclusion Matrine can up regulate the apoptosis and Bad expression of colorectal cancer cells in vitro.
