摘要
从活性污泥中筛选到一株高效絮凝剂产生菌RL-2。在1%CaCl2存在下,其发酵液对高岭土悬液具有较高的絮凝活性。根据形态及生理生化特征,利用16S rRNA部分序列分析确定该菌归属于Bacillus brevis。溶解性,茚三酮反应,Molish反应,电性试验及薄层色谱分析表明,RL-2所产高聚物为一阴离子型多糖,其结构单体为葡萄糖、半乳糖醛酸和甘露糖;由乌氏粘度法测得分子量大约为3.64×106Da。
The strain RL-2 which produced flocculating substance was screened from activated sludge.Their culture broths possessed the ability to flocculate kaolin suspension in the presence of 1% CaCl_2,which was identified to be a Bacillus brevis based on its morphological and physiological characteristics and the partial 16S rRNA sequencing method.The biopolymer produced by RL-2 was mainly composed of anionic polysaccharide,which was determined by solubility,Ninhydrin reaction,Molish reaction,electric test and TLC.It consisted of glucose,galacturonic acid and mannose with an approximate molecular size of 3.64×10~6Da.
