摘要 以木醋杆菌1.1812为出发菌株,通过单因素和正交试验,优化了木醋杆菌产细菌纤维素的发酵培养基及发酵条件。优化后的发酵培养基组成为葡萄糖8%,酵母提取物2%,柠檬酸0.3%,Na2HPO4·12H2O0.2%,KH2PO40.1%,MgSO4·7H2O0.02%,乙醇1%;发酵条件为培养基初始pH值5.4,装液量为500mL摇瓶中装100mL发酵液,发酵方式为静置培养。经验证,使用优化后的培养基配方,在上述发酵条件下,30℃培养6d,细菌纤维素的产量可达0.633g·100mL-1。 Using Acetobacter xylinum^1.1812 as primary strain, the single factor and orthogonal experiments were designed to optimize fermentation media, conditions of producing bacterial cellulose and find out a proper way of fermentation to produce more bacterial cellulose. The optimized media was obtained as follows: glucose 8% ,yeast extract powder 2%, citric acid 0.3%, Na2HPO4· 12H2O 0.2%, KH2PO40.1%, MgSO4· 7H2O 0.02%, alcohol 1%, and the proper fermentation condition as follows : pH5.4, static culture at 30℃ for 6 days. The yield is 0.633 g·100mL^-1.
