正交试验优化培养基因工程菌(E.coli pGEX)产降血压肽培养基的组成 被引量:7

摘要 通过正交试验进行摇瓶发酵,确定该菌株融合蛋白表达的最佳培养基配方,其各成分质量分数为:胰蛋白胨2.6%,酵母提取物1.9%,氯化钠0.5%,葡萄糖0.5%,另外KC l 4 mmol.L-1,MgC l210 mmol.L-1。在适宜的发酵条件下,使用优化培养基可使GST融合蛋白的表达量达到1.8 g.L-1,占总蛋白的25%。 Fermentation is a novel method of producing antihypertensive peptides (AHP). By Orthogonal test, the optimal fermentation medium for expressing the fusion protein by the engineered bacteria( E. coli/pGEX) was studied. The medium was : tryptone 2.6 %, yeast extract 1.9 %, sodium chloride 0.5 %, glucose 0.5 %, potassium 4 mmol L^-1, magnesium10 mmol L^-1 . The yield of the fusion protein was up to 1 .8 g L^-1 which was 25% of the total protein.
