摘要 目的:近来研究显示,肥胖是一种慢性低度炎症,脂肪组织能够分泌众多炎性因子,其中,白细胞介素6(IL-6)是最重要的介质之一。文中观察IL-6对人原代脂肪细胞的增殖和分化的影响。方法:用酶消化法获得人原代脂肪细胞,MTT法检测人原代脂肪细胞生长增殖的状况。分化液诱导脂肪细胞分化至产生脂滴,同时用不同浓度IL-6(0、1、5、10、50、100、1000pg/ml)干预脂肪细胞分化,油红O染色测定脂肪细胞脂滴合成状况。结果:IL-6能抑制脂肪细胞的增殖,其中5pg/ml组细胞增殖被抑制14.1%(P<0.05),10pg/ml组被抑制18.7%(P<0.05)。IL-6能够抑制细胞分化,减少脂肪的合成,其中1pg/ml组最为明显(P<0.05)。结论:IL-6能有效抑制人原代脂肪细胞的增殖,并且抑制人原代脂肪细胞从前脂肪细胞向成熟脂肪细胞的分化。 Objective : Recent studies have suggested that obedity is a state of chronic low-grade inflammation, obesity tissue could secrete kinds of inflammatory mediators. And interleukin-6 (IL-6) is one of the most important factors. Our research is to explore the effect of human IL-6 on the proliferation and differentiation of human preadipocytes. Methods : we obtained the human preadipocytes by enzyme digestion, detected the proliferation with MTT. We induced the differentiation by IL-6 at 0, 1, 5, 10, 50, 100 or 1000 pg/ml, detected the level of differentiation with oil red O. Results : The proliferation of human preadipocytes was inhibited by IL-6 at 5 pg/ml and 10 pg/ml ( P 〈 0.05 ) ; the differentiation of human preadipoeytes was also reduced by IL-6, significantly at 1 pg/ml. Conclusion:IL-6 could inhibit the proliferation and differentiation of preadipoeytes.
