Walker-256腹水大鼠单个核细胞IFN-γ、IL-4基因表达研究 被引量:1

Expression of Interferon-γ,Interleukin-4 Genes on Peripheral Blood Mononuclear Cell in Walker-256 Ascite Rat

摘要 探讨Walker-256腹水型大鼠外周血单个核细胞(peripheral blood mono-nuclear cell PBMC)中Thl和Th2类细胞因子IFN-γ、IL-4基因表达水平,进一步了解恶性肿瘤机体免疫状态的变化。建立Walker-256腹水型大鼠模型,观察其生存状态。两周后分离PBMC,用RT-PCR法检测IL-4、IFN-γ的表达水平。结果表明,腹水大鼠IL-4基因表达水平较对照组升高(分别为0.48±0.06、0.33±0.04,p<0.05),而IFN-γ基因表达水平较对照组降低(分别为0.24±0.02、0.37±0.02,p<0.05)。结论为:腹水型肿瘤大鼠Th1/Th2平衡被打破,向Th2漂移,通过检测IL-4和IFN-γ基因表达可了解肿瘤机体免疫状态。 Studing on expression level of IL-4, IFN-γ genes on peripheral blood mononuclear cell in walker-256 ascite model rat, evidence for the further study on immunoloregulation of tumor was provided. Walker-256 cell lines were used to duplicate ascite model in rat, observing their growth change. Peripheral blood mononuclear cell was collected after two weeks and quantity the expression of IL-4 and IFN-γ was tested by RT-PCR. Results shown that expression of IL-4 gene in rat is higher than that of in control group,while expression of IFN-γ gene is lower in rat group than in control group. It was conclusion the balance of Th1/Th2 is broken, immune status could be known by examing the gene expression of IFN-γ and IL-4 in ascite model.
