摘要
目的:观察Beagle犬脂肪干细胞在向成骨细胞分化的过程中富血小板血浆(platelet-richplasma,PRP)的诱导作用及其在纳米羟基磷灰石-胶原支架上的附着增殖情况。方法:酶消化法获得脂肪干细胞,分别采用含100mL/L FBS的DMEM培养液、常规成骨细胞诱导液和添加PRP的成骨细胞诱导液培养脂肪干细胞,21 d后经Von Kossa染色,观察PRP对脂肪干细胞向成骨细胞分化的诱导作用;并将脂肪干细胞与纳米羟基磷灰石-胶原共培养7d,环境扫描电镜观察细胞在支架材料上的附着情况。结果:与其他两组相比,脂肪干细胞在PRP的作用下,能生成更多的钙化结节,钙化结节细小致密,复层生长更加明显,而且脂肪干细胞能在支架材料上充分的附着伸展。结论:脂肪干细胞是良好的牙周组织工程种子细胞,PRP具有细胞因子诱导脂肪干细胞向成骨细胞的分化和增殖,纳米羟基磷灰石-胶原可以做为牙周组织工程良好的支架材料。
AIM: To investigate the effect of platelet -rich plasma (PRP) on beagle dog's adipose derived stem cells(dADSCs) when they differentiated into osteoblast, and to observe the adhesion and proliferation of dADSC on nano - HAp/Collagen. METHODS: ADSCs of beagle's dog were obtained by enzyme digestion, and cultured in DMEM with 10% FBS, osteoblast induced medium and osteoblast induced medium plus PRP respectively. The result was observed with yon Kossa staining after 21 days. dADSCs were seeded on nano -HAp/collagen scaffold material, and cell attachment was observed by environmental scanning electron microscope after 7 days. RESULTS: With the contribution of PRP, dADSCs differentiated into osteoblasts with more calcified nodules which were fine but dense, and dADSCs grew with more multilayer, dADSCs could adhere to and fully extend on nano - HAp/collagen scaffold material. CONCLUSION: dADSCs are suitable seed cells for periodontal tissue regeneration. PRP can contribute to the proliferation and differentiation of seed cells, and nano - HAp/collagen can be used as a potential scaffold material for periodontal tissue regeneration.
