摘要 目的:观察人脂肪干细胞在向成骨细胞分化过程中富血小板血浆(platelet-rich plasma,PRP)的作用,及其与支架材料纳米羟基磷灰石/胶原生物相容性的实验研究。方法:酶消化法获得脂肪干细胞,分别培养在含100ml/L FBS的DMEM培养液、成骨细胞诱导液和添加PRP的成骨细胞诱导液中,观察PRP对人脂肪干细胞向成骨细胞分化的影响。并将人脂肪干细胞与经PRP孵育的纳米羟基磷灰石/胶原复合培养7 d后,环境扫描电镜观察细胞在支架材料上附着增殖情况。结果:酶消化法能成功获得人脂肪干细胞,在PRP的作用下,脂肪干细胞能够更加有效的向成骨细胞分化,形成更多的钙化结节,结节细小致密。环境扫描电镜观察,脂肪干细胞能在支架上充分的附着伸展。结论:人脂肪干细胞的成功分离诱导分化,为牙周组织工程提供了新的种子细胞来源,PRP是促进该细胞成骨细胞分化的有效的细胞因子组合,纳米羟基磷灰石/胶原可以作为牙周组织工程的支架材料。 Objective: To investigate the effect of platelet-rich plasma(PRP) on The diffentiation of human adipose derived stem cells(hADSCs)into osteoblast,and the biocompatibility of hADSCs with a nano-HAp/collagen scaffold.Methods: hADSCs were obtained from legal abortion fetal s by enzyme digestion,and cultured in DMEM with 10% FBS,osteoblasts induced medium and osteoblasts induced medium with PRP,respectively.hADSCs were cocultured with nano-HAp/collagen which was incubiated with PRP.Cell attachment was observed by environmental scanning electron microscope after 7 days. Results: hADSCs can bs btained by enzyme digestion successfully, With the contribution of PRP, hADSCs can differentiate into osteoblast more effectively with more calcified nodules, hADSCs can adhere to and fully extend on nano - HAp/collagen scaffold under environmental scanning electron microscope. Conclusion: hADSCs' isolation and differentiation offered a new chioce of seed cells for periodontal tissue engineering. PRP was an effective cell factor for bADSCs'differentiation into osteoblasts. Nano - HAp/collagen could be used as a potential scaffold in periodontal tissue regeneration.
