摘要
目的采用TLC法鉴别桃儿七果实药材中的鬼臼毒素并采用HPLC法测定含量。方法色谱柱为Phenomenex LunaC18(2)(150mm×4.6mm,5μm);流动相为乙腈-0.2%磷酸溶液(38:62);检测波长290nm;流速1.0ml·min-1;柱温32℃。结果各批桃儿七果实药材TLC色谱中均能检出鬼臼毒素;鬼臼毒素与其他色谱峰分离良好,进样量0.26—1.30μg呈良好线性关系(r=0.9999),平均回收率为97.84%,RSD=1.66%(n=6)。结论所建方法简便快捷,结果可靠,可控制桃儿七药材的质量。
OBJECTIVE To establish the TLC identification and quantitative determination methods of the fruit of Sinopodophyllum Royle. METHODS TLC and HPLC were adopted to identify and determine the furit of Sinopodophyllum Royle. The chromatographic conditions were : Phenomenex Luna C18 ( 2 ) ( 150 mm × 4.6 mm ,5 μm) column as analytic column, acetonitrile - 0. 2% Ha PO4 water solution (38 : 62) as mobile phase; detection wavelength at 290 nm;column temperature at 32~C ;flow rate at 1.0 ml. min-1. RE- SULTS Podophyllotoxin was identified by TLC and determined by HPLC. The liner range of podophyllotoxin was 0. 26 - 1.3 μg ( r = 0. 9999). The average recovery was 97. 84% with RSD of 1.66% ( n = 6). CONCLUSION These methods are simple, accurate, reproducible and can be used to improve the quality control of this drug.
