摘要 建立高效液相色谱法测定4种八角莲中鬼臼毒素含量的方法,考察不同溶剂及提取方法对含量测定结果的影响。方法Hewlet-Packard1100高效液相色谱仪,WhatmanPartiShereC18色谱柱,流动相:甲醇-0.1mol/L磷酸氢二钾(50∶50),检测波长:290nm。样品用80%的甲醇水溶液超声波振荡提取结果标准曲线方程Y=0.225+438.95X,r=0.9997(n=3)。结论超声波法优于传统的回流提取法,本方法简便快速,准确度高。 PURPOSE To develop a method for determination of podophyllotoxin in Dysosma plants. Solvents and methods used to extract was tested. METHODS A high performance liquid chromatography method was used with Whatman PartiShere C 18 . The mobile phase was methanol 0.1mol/L K 2HPO 3(pH≈7.5)(50∶50). Flow rate was 1.0ml/min. Detection wavelength was at 290nm. The plants were extract by ultrasonic method with 80% methanol. RESULTS The linear range was from 2.6 to 13.0μg/ml. The standard curve equation was Y =0.225+438.95 X and the relevant coefficient was 0.9997( n =3). CONCLUSIONS This method was proved to be accuricy and simple.
