摘要
目的研究路边青与穿心莲配伍使用对脱水穿心莲内酯(DAP)的小鼠血液药动学性质的影响。方法小鼠灌胃穿心莲总内酯干浸膏和复方穿心莲干浸膏药液后于不同时间点取血,血样经前处理后用反相高效液相色谱分离、测定脱水穿心莲内酯含量,采用3P97药动学软件对数据进行处理。结果小鼠灌胃穿心莲总内酯干浸膏和复方穿心莲片干浸膏后,脱水穿心莲内酯的血药浓度动态变化规律均符合一室开放模型。路边青与穿心莲配伍使用可导致脱水穿心莲内酯的吸收慢、消除快、生物利用度低。结论穿心莲与路边青配伍使用是否科学,值得深入研究。
OBJECTIVE To study the pharmacokinetics of dehydroandrographolide in Andrographis & Clerodendron compatible preparation in mice. METHODS After oral administration of total andrographolide and Andrographis & Clerodendron compatible preparation to mice by gavage respectively, blood samples were collected at different time points. The contents of dehydroandrographolide were determined. The mean plasma concentration-time profile of dehydroandrographolide was processed by 3P97 pharmacokinetic software. RESULTS The mice plasma concentration-time curves of dehydroandrographolide, after administered total andrographolide and Andrographis & Clerodendron compatible preparation by gavage respectively, were fitted to one-compartment open model. The use of Andrographis together with Clerodendron delayed the absorption, increased the clearance, and decreased the bioavailability ofdehydroandrographolide. CONCLUSION The compatibility ofAndrographis & Clerodendron should be investigated in the future.
