摘要
目的探讨痛泻要方煎剂对肠易激综合征(IBS)内脏高敏感性模型大鼠的肠道肥大细胞的影响。方法选用清洁级新生SD大鼠,采用结肠慢性刺激法加夹尾刺激法造模。造模成功后,将实验动物分组为空白对照组、模型对照组、痛泻要方煎剂低剂量组(低剂量组)、痛泻要方煎剂中剂量组(中剂量组)、痛泻要方煎剂高剂量组(高剂量组)、奥替溴铵片组。空白对照组和模型对照组灌胃生理盐水,其余各组分别灌胃相应药物,每天两次,连续30d。结果模型组大鼠回盲部肥大细胞数量较对照组明显增加,痛泻要方煎剂高剂量组与奥替溴铵片同样能降低模型大鼠回盲部肥大细胞数目,与模型组比较有显著性差异。结论痛泻要方煎剂能抑制模型大鼠肥大细胞脱颗粒,减少肥大细胞数量,可能是其作用机制之一。
Objective To explore the impacts of Tongxieyaofang Decoction( the decoction of invaluable formula for pain and diarrhea) on intestinal mast cells in rat model with visceral hypersensitivity in irritable bowel syndrome(IBS). Methods Sanitary-class, neonatal SD rats were optioned and chronic rectal irritation and tail - clipping stimulation was adopted for modeling. After successfully modeled, the experimental animals were divided into blank control group, model control group, low dosage of Tongxieyaofang Decoction group(low-dosage group), moderate dosage of Tongxieyaofang Decoction( moderate-dosage group), high dosage of Tongxieyaofang Decoction(high-dosage group)and Otilonium Bromide Tablets group. Gastric per- fusion with physiological saline was provided in blank control and model control groups. Gastric perfusion with corresponding drugs was provided in the rest groups, twice a day, continuously for 30 days. Results The counts of mast cells in ileocecum in model group were increased compared with those in control groups. Tongxieyaofang Decoction of high dosage and Otilonium Bromide Tablets reduced the counts of masts cells in ileocecum in model rats, indicating significant difference compared with that in model group. Conclusion Tongxieyaofang Decoction can inhibit mast cell degranulation in model rats and reduce the counts of mast cells,which is probably one of the mechanisms on its efficacy.
