摘要
目的探讨基质金属蛋白酶-13(MMP-13)及MMP-13 mRNA在人肝细胞肝癌(HCC)中的表达和意义。方法分别应用免疫组化和原位杂交方法检测32例HCC患者手术切除的癌组织及其癌旁组织中的MMP-13蛋白和MMP-13 mRNA,分析临床病理资料与MMP-13的关系。结果HCC组织中MMP-13和MMP-13 mRNA阳性表达率为93.75%(30/32)和90.63%(29/32),癌旁组织中的阳性表达率为71.88%(23/32)和59.38%(19/32),两组比较差异有统计学意义(P<0.01)。MMP-13在肿瘤中的表达与肿瘤组织分化程度、有无门静脉癌栓和包膜侵犯密切相关(P<0.05)。结论MMP-13及MMP-13 mRNA在HCC组织中的表达明显高于癌旁组织,且与组织分化、门脉癌栓等临床病理有关,提示MMP-13在肿瘤的生长过程中发挥一定作用,可能与HCC的侵袭和转移有关。
Objective To study the expression and significance of matrix metalloproteinase-13 (MMP-13) and MMP-13 mRNA in human hepatocellular carcinoma (HCC). Methods MMP-13 and MMP-13 mRNA in HCC and tumor-adjacent tissues of 32 cases were measured by immunohistochemical staining and in situ hybridization respectively. Meanwhile the relationship between the expression of MMP-13 and clinical pathology was analyzed. Results The positive rates of MMP-13 and MMP-13 mRNA were 93. 75% (30/32)and 90.63% (29/32)in HCC, and 71.88% (23/32)and 59.38%(19/32)in tumor-adjacent liver tissues. The expressions of MMP-13 and MMP13 mRNA were significantly higher in HCC than those in tumor-adjacent tissues (P〈0. 01). The positive expression rate of MMP-13 was higher in the cases of poorly -differentiation and PVTT than that of well-or middle differentiation and no PVTT in HCC (P〈0.05). Conclusion MMP-13 and MMP-13 rnRNA are over-expressed in HCC tissues, indicating that MMP13 may have important roles in the development Of HCC.
