Effects of Angiopioetin-2, VEGF and bFGF Expression on Hepatocellular Carcinoma Angiogenesis

摘要 【目的】研究血管生成素2(Ang2)在人肝细胞癌(HCC)组织中的表达,并探讨其与HCC组织中血管内皮生长因子(VEGF)、碱性成纤维生长因子(bFGF)和微血管密度(MVD)的关系。【方法】采用免疫组化和逆转录聚合酶链反应(RTPCR)的方法,对38例HCC的癌组织及癌旁非肿瘤组织,7例正常肝组织中的Ang2、VEGF、bFGF和MVD的表达进行检测;同时研究了肝癌组织中Ang2的表达与临床病理关系。【结果】Ang2在正常肝组织无表达,癌旁组织呈弱表达(39.47%),在大部分癌组织呈强表达(60.53%)。Ang2表达与VEGF、bFGF和MVD表达有明显相关性(r分别为0.857、0.706和0.820,P<0.01)。Ang2表达与HCC的血管侵犯、肿瘤的转移及肿瘤的分级有关(P<0.05)。【结论】Ang2、VEGF和bFGF在HCC血管形成和进展中具有协同作用。Ang2、VEGF、bFGF及MVD可作为HCC生物学行为恶化的一项评估指标。 [Objective]To study the expressions of angiopioetin-2( Ang-2 ) ,vascular endothelial growth factor(VEGF) , basic fibroblast growth factor(bFGF ) and mierovascular density(MVD) in human hepatocellular carcinoma (HCC), and their relationship to the angiogenesis and progression of hepatoeellular carcinoma (HCC). [Methods]The Ang-2 expression was examined in thirty-eight matched sets of HCC specimens and adjacent liver, and sevent normal liver tissues, by immunohistochemistry and reverse transcription- polymerase chain reaction (RT-PCR). The relationship among Ang-2,VEGF, bFGF and MVD expression and the clinical pathological characteristics of HCC were studied. [Results]Ang-2 was not expressed in normal liver tissues, weakly expressed in adjacent nontumorous livers(39.47% ), but strong expressed in HCC(60.53%). The expression of Ang-2 was significantly correlated with VEGF , bFGF and MVD (r1 = 0. 857, r2 = 0. 706, r3 = 0. 820, P〈0. 01). A high Ang-2 ratio in HCC was associated with tumor vascular invasion, metastasis and differentiation( P 〈0.05). [Conclusion]Ang-2 gene plays an important role in the carcinogenesis and progression of HCC. Ang-2.VEGF and bFGF may play an important role in a synergistic manner in HCC. Ang-2,VEGF, bFGF and MVD may serve as tumor markers to evaluate the biological behavior of HCC.
