摘要
[目的]研究三氯化铝(AlCl3)侧脑室注射对小鼠空间学习和记忆的影响。[方法]取经过跳台实验、避暗实验和Morris水迷宫实验筛选的正常C57BL小鼠40只随机分为4组,即对照组、AlCl3组、Al3++生理盐水组和Al3++双左旋2-氨基3-磷羧基丙酸(Al3++DL-AP3)组,每组10只;采用侧脑室注射AlCl3,制成阿尔茨海默病(Alzheimer’sdisease,AD)小鼠模型,再应用跳台实验、避暗实验和Morris水迷宫实验判断给药前后小鼠的空间学习和记忆能力。[结果]AlCl3模型组跳台实验的潜伏期较对照组明显缩短(P<0.05),跳台实验及避暗实验的错误次数均明显增多(P<0.05),Morris水迷宫实验中定位导航实验潜伏期较对照组明显增加(P<0.05),空间探索试验的潜伏期缩短(P<0.05)。与Al3++生理盐水组相比,Al3++DL-AP3组跳台实验潜伏期增加(P<0.05),跳台实验及避暗实验的错误次数均减少(P<0.05),Morris水迷宫实验中定位导航实验潜伏期明显缩短(P<0.05)。[结论]AlCl3侧脑室注射可导致小鼠空间学习和记忆障碍,DL-AP3对这种空间学习和记忆障碍有一定的预防和治疗作用。
[ Objective ] To investigate the effects of spacial learning and memory of Alzheimer' s disease( AD )model in C57BL mouse induced by aluminum chloride(AlCl3)lateral cerebral ventricle injection. [ Methods ] After successfully trained by step through test, step down test and Morris water maze ( MWM ), 40 mice were randomly and evenly divided into 4 groups: control group, A1Cl3 group, A1Cl3+normal saline group (Al^3+ +NS) and AlCl3+DL-2-Amino-3-phosphonopropionic group (Al^3+ +DL-AP3). AlCl3 was injected into lateral cerebral ventricle and drug pre-treatment was followed for one week. Spatial learning and memory ability were evaluated by step through test, step down test and MWM after 21d drug therapy. [Results] The latent period of step down test was significant decreased(P〈0.05), and the error number of step down test and step through test were significant increased(P〈0.05)as compared with the control group, in MWM the time of place navigation significant increased( P 〈 0.05 ), the time of spatial probe significant shortened( P 〈 0.05 ). Compared with the Al^3+ +NS group, the time of place navigation was significantly shortened in the Al^3+ +DL-AP3 group(P〈0.05). It also significantly prolonged(P〈0.05)the latent period of step through test, and significantly decreased the error number(P〈0.05)of step down test and step through test. [Conclusion] The ability of spatial learning and memory of mice were descended obviously by AlCl3 lateral ventricle injection. The DL-AP3 could improve the ability of spatial learning and memory.
