摘要
目的:观察碱性成纤维细胞生长因子(bFGF)对糖尿病大鼠心肌缺血再灌注(I/R)损伤的影响。方法:48只大鼠分为正常I/R+生理盐水组(CN)、正常I/R+bFGF组(Cb)、糖尿病I/R+生理盐水组(DN)、糖尿病I/R+bFGF组(Db)。以腹腔注射链脲佐菌素(STZ)法建立糖尿病模型,以结扎冠状动脉左前降支法建立I/R模型。每组6只颈动脉取血,离心得血清,以丙酮酸二硝基苯腙显色法检测乳酸脱氢酶(LDH)活性,硫代巴比妥酸(TBA)法检测丙二醛(MDA)含量。颈动脉取血后取心脏做心肌组织石蜡包埋,免疫组化染色检测第Ⅷ因子的表达,计算心肌血管数量。每组剩余6只取心脏做TTC染色,计算心肌梗死面积。结果:I/R后,Cb组和Db组心肌血管数高于对应的CN组和DN组,LDH活性和MDA含量低于CN组和DN组;DN组心肌血管数低于CN组,DN组LDH活性和MDA含量高于CN组;Db组心肌血管数低于Cb组,而LDH活性和MDA含量高于Cb组。结论:糖尿病大鼠I/R后心功能受损和梗死程度比正常大鼠严重,补充bFGF可以改善心功能和减少心肌梗死面积,但疗效不如非糖尿病组。
Objective: To evaluate the effects of bFGF on I/R induced myocardial injury in diabetic rats. Methods: Forty-eight rats were randomly divided into four groups: non-diabetic I/R group (CN group), non-diabetic rats treated with bFGF group (Cb group), diabetic I/R group (DN group), diabetic rats treated with bFUF group (Db group). Diabetes was induced by intraperitoneal injecting STZ, and I/R by ligating left anterior descending coronary artery. Blood drawn from carotid artery was collected in six cases in each group, then the hearts were harvested for paraffin-embedded. The activity of lactate dehydrogenase (LDH)and contents of malondialdehyde (MDA) were measured. Ⅷ factor was detected in six cases in each group with immunohistochemistry.The residual six cases hearts were harvested for TTC staining to calculate infarcted myocardial area. Results: After I/R, the number of capillary was significantly higher in Cb group and Db group than those in CN group and DN group. The activity and content of LDH and MDA were significantly lower than those in CN group and DN group. The number of capillary was lower in DN group than that in C group, while the activity and content of LDH and MDA were higher than those in C group. The number of capillary was lower in Db group than that in Cb group, while activity and content of LDH and MI)A were higher than those in Cb group. Conclusion: Diabetic rat hearts are more susceptible to I/R induced cardiac dysfunction and infarction degree, bFGF can improve the recovery of cardiac function and reduce the severity of I/R induced infarction in both non-diabetic rats and diabetic rats, but the therapeutic effects are less effective in diabetic rats than those in non-diabetic rats.
