Effects of Sea Buckthorn Oil and Schisandrin B on Sub-Chronic Nephrotoxicity Induced by Cadmium in Rats

摘要 目的探讨沙棘油(sea buckthom oil,SBO)和五味子乙素(schisandrin B,Sch B)对亚慢性镉中毒致大鼠肾脏氧化损伤的影响。方法将健康清洁级Wistar大鼠按体重随机分为对照组(生理盐水)、单纯镉染毒组、CdCl_2+SBO染毒组和CdCl_2+Sch B染毒组,每组8只。对照组皮下注射生理盐水,单纯镉染毒组、CdCl_2+SBO染毒组和CdCl_2+Sch B染毒组均皮下注射6μmol/kg的CdCl_2,每天1次,注射容量为5 ml/kg,连续染毒6周;第7、8周,对照组和单纯镉染毒组灌胃生理盐水,灌胃容量为5 ml/kg;CdCl_2+SBO染毒组和CdCl_2+Sch B染毒组分别灌胃5 ml/kg的SBO和8 mg/kg的Sch B。染毒后,收集大鼠18 h尿液,然后处死大鼠,取肾组织,测定大鼠尿中乳酸脱氢酶(LDH)、N-乙酰-β-D-氨基葡萄糖苷酶(NAG)活力及尿蛋白、尿肌酐含量,肾皮质中谷胱甘肽(GSH)、丙二醛(MDA)含量及超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(CSH-Px)活力。结果与对照组比较,单纯镉染毒组尿中LDH、NAG活力和尿蛋白含量、肾组织中GSH、MDA含量升高,SOD和GSH-Px活力下降,差异均有统计学意义(P<0.01)。与单纯镉染毒组比较,CdCl_2+SBO染毒组和CdCl_2+Sch B染毒组大鼠尿中LDH、NAG活力和尿蛋白含量、肾皮质中GSH、MDA含量降低,SOD和GSH-Px活力升高,差异均有统计学意义(P<0.05或P<0.01)。结论SBO和Sch B对亚慢性染镉致肾氧化损伤的恢复具有一定的促进作用。 Objective To study the effects of sea buckthorn oil(SBO) and schisandrin B(Sch B) on sub-chronic oxidative renal injury induced by cadmium(Cd). Methods Thirty-two adult Wistar rats were randomly divided into four groups. Control group was injected subcutaneously with 0.9% NaC1. Cadmium group and SBO, Sch B pretreatment groups were injected subcutaneously with cadmium chloride(CdCl2) 6 umol/kg, for up to 6 weeks. Then the two pretreatment groups were respectively given SBO 5 ml/kg and Sch B 8 mg/kg by gavage for 2 weeks. Control group and cadmium group were given 0.9% NaCl at the same time. The urine and kidney cortex samples were collected in the end of the 8th week. The activity of lactate dehydrogenase(LDH), n-acetylglucosaminidase (NAG) activity and protein contents in urine were determined. Glntathione (GSH) and malonydialdehyde (MDA) contents in renal cortex as well as superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) activity were measured. Results Compared with the control group, the activity of LDH and NAG, the protein contents in urine, the contents of GSH and MDA in the kidney cortex obviously increased. Adversely, the activity of SOD and GSH-Px decreased significantly in the group treated with Cd alone(P〈0.01). The contents of GSH and MDA increased sharply, the activity of SOD and GSH-Px in the kidney cortex decreased significantly both in the SBO group and Sch B group (P〈0.05 or P〈0.01). Compared with the Cd group, the activity of LDH and NAG, the protein contents in urine significantly reduced, GSH and MDA contents declined sharply, SOD and GSH-Px activity in the kidney cortex increased significantly in the SBO group as well as the Sch B group(P〈0.05 or P〈0.01). Conclusion SBO and Sch B present some protective effects on Cd induced sub-chronic oxidative damages in the kidneys.
