乌梅散对镉致小鼠肝肾损伤的保护作用

PROTECTION EFFECT OF WUMEISAN ON CADMIUM EXPOSURE IN MICE LIVER AND KIDNEY DAMAGE

摘要 目的研究乌梅散对染镉所致小鼠肝脏和肾脏损伤的保护作用。方法昆明小鼠随机分为四组,每组10只,空白对照组、镉染毒组、乌梅散高剂量组和低剂量组。低剂量组灌胃给予乌梅散50mg/(kg·d),高剂量组100mg/(kg·d),各组每天灌胃一次,连续30d。镉染毒组和乌梅散组腹腔注射氯化镉3mg/(kg·d)染毒,连续染毒10d。第三十天,测定小鼠血清天冬氨酸转氨酶(AST)和丙氨酸转氨酶(ALT)活力,测定血清、肝肾组织中的超氧化物歧化酶(SOD)活力和丙二醛(MDA)含量。观察肝组织和肾组织病理学变化。结果与空白组相比,镉染毒组小鼠血清中AST、ALT水平明显上升(P<0.05),血清、肝肾组织中SOD活力显著减低(P<0.05),而MDA含量明显升高(P<0.05)。与镉染毒组相比,乌梅散低、高剂量组小鼠血清中AST、ALT活力明显降低(P<0.05),血清、肝肾组织中SOD活性显著升高(P<0.05),MDA含量明显降低(P<0.05)。病理结果显示,镉染毒组小鼠肝细胞肿胀,有片状细胞环,肾近曲小管可见管腔形态不完整;乌梅散高剂量组肝细胞损伤程度有所减轻。乌梅散高剂量组小鼠近曲小管受损程度减轻。结论乌梅散对镉染毒所致的小鼠肝肾损伤有保护作用。 Objective To study the protective effects of Wumeisan on cadmium induced liver and kidney damage in mice.Methods Kunming mice was divided into 4 groups randomly,control group,cadium exposed group and Wumeisan low dose Wumeisan group and high dose Wumeisan group.The mice of control group and cadmium exposure were feeding on pure water,the low dose group were given Wumeisan50mg/(kg·d),the high dose group were given 100mg/(kg·d)for 30 days.Control group and cadmium exposure were ip cadmium chloride simutaneously,with the dose of 3mg/(kg·d)for 10 d.After given dose,the activity of alanine aminotransferase(AST)and aspartate aminotransferase(ALT)were detected in serum,and the homogenate superoxide dismutase activity and content of malondialdehyde were determined in liver,kidney and serum.Liver and kidney morphology were observed.Results Compared with the control group,the AST and ALT activity of cadmium exposure group were significantly increased(P〈0.05);Compared with cadmium exposure group,the AST and ALT activity of Wumeisan group were decreased significantly(P〈0.05).The activity of SOD in the mice serum,liver and kidney was increased,but MDA content was significantly reduced(P〈0.05).Pathology showed that liver cells were swelling,in cadmium exposure group,with a sheet necrocytosis,and with a incomplete mophology in lumen of proximal convoluted tubule in kidney.But in high dose Wumeisan group,hepatocellular injury was alleviative and the damage of proximal convoluted tubule was also decreasing.Conclusion Wumeisan has a protective effect on cadmium exposure damaged liver and kidney.
