摘要
目的研究川楝子致小鼠肝毒性的时效、量效关系。方法小鼠灌服不同剂量川楝子,测定其LD50。另取小鼠按不同时间点或不同剂量分组,检查血清ALT,AST,计算肝、脾、胸腺脏器指数,并观察肝组织形态变化。结果小鼠灌服川楝子的LD50为200 g.kg-1。ALT,AST在给药后1 h达到高峰,24 h可恢复近正常值。与正常组比较,川楝子81,166 g/kg剂量组对肝组织产生明显损伤,且随着剂量增大,ALT,AST升高显著(P<0.05或P<0.01)。结论小鼠单次灌服一定剂量川楝子可造成急性肝损伤甚至死亡,并显示有毒性时效、量效关系。
Objective To study the time - effect relation and dose - effect relation of hepatotoxicity induced by Fructus Toosendan in mice. Methods The medial lethal dose ( LD50 ) of mice was calculated after different doses of Fructus Toosendan was given orally. Then the mice were divided into different groups according to different doses or time, and the contents of serum alanine aminotransferase(ALT) , aspartate aminotransferase(AST) were investigated, the thymus , liver and spleen were weighed , and the liver tissue was studied with histopathological to observe the degrees of inflammatory activity and fibrous hyperplasia. Results The LD50 of Fructus Toosendan was 200 g/kg, 95 % confidence level of which was 192 - 212 g/kg, ALT, AST in serum reached its peak after 1 hour, which recovered closely to normal value after 24 hours. Compared with control group, hepatic tissues of the high and medium dose groups were damaged significantly, and the contents of ALT and AST in serum were increased significantly ( P 〈 0.05 or P 〈 0.01 ). Conclusion When the mice were fed with a certain dose of Fructus Toosendan relation for single time, it will be induced acute liver toxicity and even death. Time - dose - toxicity effects would be indicated of Fructus Toosendan.
