摘要
目的探讨生长抑素类似物奥曲肽诱导人胃癌细胞株SGC-7901细胞凋亡的机制。方法将胃癌细胞分为6组:不加药组(对照组)、奥曲肽0.1mg/L组、0.2mg/L组、0.5mg/L组、1.0mg/L组、2.0mg/L组。RT-PCR扩增后,凝胶电泳观察hTERT表达情况,AO染色、荧光显微镜下观察细胞凋亡情况。结果胃癌细胞中有hTERT表达,用RT-PCR技术扩增hTERT基因片段,经2%琼脂糖凝胶电泳显示获得了145bp的扩增条带,随着奥曲肽浓度的增大,hTERT表达逐渐减少,凋亡细胞数逐渐增多。结论奥曲肽诱导胃癌细胞凋亡的机制可能与抑制胃癌细胞中hTERT的表达有关。
Objective To explore the mechanism of somatostatin analogs octreotide inducing cell apoptosis in human gastric cancer cell line SGC-7901. Methods Different concentrations of octreotide(0.1,0.2,0.5,1.0,2.0 mg/L) were added into the human gastric cells in octreotide groups, respectively, but no medicine was added in positive control group. The expression of hTERT was determined by reverse transeriptase polymerase chain reaction (RT-PCR). The cell apoptosis was observed under fluorescence microscope. Results The hTERT expressed in the gastric cancer cells. The hTERT gene fragment was amplified by RT-PCR, and 145 bp amplification product was detected by 2% agarose gel electrophoresis. With the increase of octreotide concentration, the expression of hTERT reduced gradually and the number of apoptosis cells increased gradually. Conclusion Octreotide could induce the gastric cancer cell apoptosis and inhibit the expression of hTERT in the gastric cancer cell. The apoptosis might be related to the inhibition of hTERT expression in gastric cancer cell.
