2004~2006年江西省甲3亚型流感疫苗预防效果的基因分析 被引量:3

摘要 [目的]了解江西省2004—2006年分离的甲3亚型流感病毒株HA1基因变异特征,分析WHO甲3亚型疫苗推荐株对江西省甲3亚型流感的预防效果。[方法j采集监测医院和疑似流感疫情的流感样病例鼻咽拭子标本进行流感病毒分离,对分离到的H3型流感病毒进行核酸提取,采用逆转录-聚合酶链反应(RT—PCR)扩增病毒HA1基因后进行核苷酸序列测定,用DNAStar5.0、BioEdit(Version5.0)、Mage生物软件对测序结果进行分析处理,推导出氧基酸序列,进行基因特性分析。[结果]21株甲3亚型流感病毒HA1区域核苷酸序列长度均为960bp.未发现核苷酸的丢失和插入,平均点突变率为1.09%。2004年甲3亚型流感病毒分离株与当年WHO疫苗推荐株HA1区相比较.不同毒株氨基酸的同源性是97.8%-98.8%,抗原决定篱区的氧基酸变异数平均为3.17个,但有毒株已出现在2个抗原决定簇区4个氨基酸替换。2005年甲3亚型流感病毒分离株与当年疫苗推荐株HA1区相比较,不同毒株氨基酸的同源性是98.4%~99.1%.抗原决定簇区的氨基酸变异数平均为2.0个。2006年甲3亚型流感病毒分离株与当年疫苗推荐株的HA1区相比较,不同毒株氡基酸的同源性是98.1%~98.4%.在2个抗原决定簇区的氨基酸变异数平均为5.71个。f结论12004年、2005年和2006年分别根据WHO推荐疫苗株生产的疫苗,对我省2004年度的甲3亚型流感总体上有较好的预防效果,但毒株的变异在增强;对2005年度的甲3亚型漉感上有良好的预防效果;对2006年度的甲3亚型流感预防效果不够理想。 [Objective] To know the mutation on influenza viruses subtype A3 in Jiangxi province and the differences on HA1 regions between isolation strains and vaccine strains of influenza viruses subtype A3 from 2004 to 2006, and analyze the protective effect of the vaccine strain reeonunended by WHO. [ Methods] The specimens of nasopharyngeal swabs were collected from influenza like cases of monitor hospitals and influenza epidemic situation. RNA was extracted from the influenza viruses subtype A3 isolated from the specimens, then their HA1 genes were amplified by reverse transcription polymerase chain reaction (RT-PCR) . The PCR products were sequenced and the data obtained were analyzed with the software DNAStarS.0, BioEdit (Version5.0) and Mage. [Results] The nucleotides of HA1 region of 21 strains were all 960 bp, there were no deletion and insertion in the nucleotides. Accordingly, the average percent of point mutation was 1.09% .The strain isolated in 2004, in 2005, and that in 2006 had respectively 97.8%-98.8%, 98.4%-99.1%, and 98.1%-98.4% identity in amino acid sequence compared with vaccine strain recommended by WHO at that time. There were respectively 3.17, 2.0, and 5.71variant amino acids found locating in 2 antigenic determinant sites from 2004 to 2006. But there was 4.0 variant amino acids found locating in 2 antigenic determinant sites in some isolation strain in 2004. [Conclusion] The vaccine produced by the influenza subtype A3 vaccine strain recommended by WHO has good protective effect to the subtype A3 influenza in general in Jiangxi Province in 2004, but increasing variation on HA1 region of influenza subtype A3 virus isolated is found. The vaccine produced by the influenza subtype A3 vaccine strain recommended by WHO in 2005 has better protective effect, but that in 2006 has worse protective effect.
