摘要
对根癌农杆菌介导的水稻遗传转化受体系统的几个重要因素进行试验。得到157个抗性愈伤组织。对于滇优粳3号和滇优粳5号,4mg/L的2,4-D和0.2mg/L的6-BA搭配,蔗糖浓度为20~30g/L时比较有利于愈伤组织形成。滇优粳3号和滇优粳5号的愈伤组织对潮霉素较敏感,30~50mg/L的靓霉素浓度为有效的筛选浓度。
Two rice varieties, Dianyoujing 3 and Dianyoujing 5, were tested to study some important factors affecting rice genetic transformation system mediated by agobacterium tumefaciens and 157 callus with Hyg resistance were obtained. The results showed that the callus induction rate of Dianyoujing 3 and Dianyoujing 5 was 72.50% and 71.21% under the culture conditions of MS+ 4 mg/L 2, 4-D, 0.2 mg/L 6-BA and 20~30 g/L cane sugar respectively. The optimum hygromyein concentration for selecting callus with Hyg resistance was to 30~50 mg/L in Dianyoujing 3 and Dianyoujing 5.
