摘要
目的:研究玉泉丸对盐酸二甲双胍在糖尿病大鼠体内药动学过程的影响。方法:糖尿病模型大鼠灌胃给予玉泉丸7 d后,给予盐酸二甲双胍,给药后于不同时间采集血样,用高效液相色谱方法测定血药浓度,计算药动学参数。结果:二甲双胍组、联合用药组的药动学参数Cmax分别是18.95,21.76 mg.L-1;t1/2分别是1 069.8,1 767.4 min;CL/F分别是0.013,0.008 L.min-1.kg-1;AUC分别是10 042.1,10 712.2 mg.L-1.min-1。结论:盐酸二甲双胍在糖尿病大鼠体内药动学过程符合一室模型,玉泉丸对盐酸二甲双胍在糖尿病大鼠体内的药动学有显著影响。
Objective: To study effects of Yuquan pills on the pharmacokinetics process of mefformin hydrochloride in diabetic rats. Method: After administration Yuquan pills 7 day to the diabetic rats, the metformin hydrochloride was orally administrated, then the blood samples were collected at different time. The concentrations of mefformin hydrochloride in plasma were determined by HPLC method and the pharmacokinetic parameters were calculated. Result: The pharmacokinetie parameter Cmax of the controlling group and the testing group were respectively, 18. 95, 21.76 mg·L^-1 ; t/2 were 1 069. 8, 1 767.4 rain, respectively; CL/F were 0. 013, 0. 008 L ·min^-1· kg-1 ; AUC were 10 042. 1, 10 712. 2 mg · L^-1 . min-1 respectively. Conclusion: The pharmacokineties process of metformin hydrochloride in diabetic rats fits one-compartment model. Yuquan pills has a significant effect on the pharmacokinetics of metformin hydrochloride in diabetic rats.
