中国部分家养山羊mtDNA D环区遗传多样性与进化 被引量:6

Genetic diversity and origin of mitochondrial DNA D-loop region for some domestic goat breeds of China

摘要 对我国10个家养山羊品种140只个体的mtDNAD-loop区进行测序分析,测得结果:整个D-loop区为1211~1213bp,共检测到84种单倍型,171个多态位点。其中核苷酸多样性(Nucleotide diversity,Pi)为:0.02063±0.00225,单倍型多样性(Haplotypegene diversity,Hd)为:0.988±0.003,平均核苷酸差异数(Average number of nucleotide differences)k为:24.896。表明我国家养山羊品种遗传多样性丰富。通过构建NJ网络进化树,得出中国家养山羊主要分为两大支系,并且其中一支与角骨羊(Capra aegagrus)聚在一起,而旋角野山羊(Capra falconeri)单独聚为一支,说明角骨羊对中国家养山羊贡献较大。 The complete sequences of mitochondrial DNA D-loop of 140 individuals in 10 Chinese goat (Capra hiruc) breeds were analyzed by DNA sequencing technology. The results showed that the length of mtDNA D-loop in Chinese goats was 1211-1213 bp. There were 84 different haplotypes and 171 polymorphic sites. The mean nucleotide diversity (Pi) and haplotype diversity (Hd) were 0.02063 ±0.00225 and 0.988± 0.003, respectively, and the average number of nu- cleotide differences (k) was 24.896. The results showed an abundant genetic diversity in domestic goats of China. The NJ tree indicated that there were two main branches in Chinese domestic goats, thereinto, one branch was clustered with Capra aegagrus, and Caprafalconeri was clustered alone, which indicated that Capra aegagrus had more contribution to Chinese domestic goat breeds.
