阿昔洛韦口腔缓释膜的制备及其体外释放 被引量:7

摘要 目的:以魔芋葡甘聚糖和黄原胶两种天然多糖为基质,制备新型阿昔洛韦口腔缓释膜,用于疱疹性口腔溃疡局部给药。方法:1%(W/W)魔芋葡甘聚糖和1%(W/W)黄原胶溶液按体积比7∶3配制成混合凝胶45ml,加入0.4ml甘油,0.5ml吐温-80,再加入已溶解于5ml水中的阿昔洛韦0.538g,取30ml胶液在5cm×8cm无菌玻璃槽中干燥成膜,制成直径为1.5cm的膜片。通过紫外分光光度法进行阿昔洛韦含量和体外累积释放度测定。结果:所得药膜的阿昔洛韦含量为(13.87±0.34)mg/片,在30min时体外累积释放度为21.56%,而4.5h时达到最大释放,为92.25%。结论:魔芋葡甘聚糖和黄原胶复配凝胶基质的阿昔洛韦口腔膜剂,具有较好的缓释效果,可望进一步深入开发。 Objective: Konjac glucomannan and xanthan gum as matrix to prepare Aciclovir Oral Sustained Release Membrane for oral herpes ulcer treatment. Methods: 1% (W/W) of konjac glucomanan and 1% (W/W) of xanthan were mixed at the ratio of 7∶3 (V/V) to form 45 ml of mixed gel, 0.4 ml of glycerin and 0.5 ml of Tween-80 were added, 0.538 g of aciclovir dissolved in 5 ml of water and also was added. 30 ml of mixed gel was pulled on the surface of a 5 cm×8 cm sterilized glass and dried to form membrane, then prepared into some small membranes with diameter of 1.5 cm. The content of aciclovir and the in vitro cumulative release were determined by UV spectrography. Results: The aciclovir mean content of each piece of resulting membrane was (13.87±0.34) mg, the in vitro cumulative release was 21.56% at 30 min, reach the maximum of 92.25% at 4.5 h, and could maintain 3-7 h. Conclusion: Konjac glucomannan and xanthan mixture gel-based Aciclovir Oral Sustained Release Membrane has good sustained release characteristics, and worth to be developed in the future.
