摘要
目的探讨关节腔注射黄原胶对兔膝骨关节炎关节滑液中白介素1β(IL-1β)、肿瘤坏死因子α(TNF-α)、前列腺素E2(PG E2)和一氧化氮(NO)含量的影响。方法用木瓜蛋白酶建立兔膝骨关节炎模型。造模后将兔随机分3组,分别于膝关节腔注射生理盐水、玻璃酸钠和黄原胶。采用酶联免疫吸附法检测关节滑液中IL-1β、TNF-α和PG E2的含量,采用硝酸酶还原法检测NO的含量。结果与生理盐水组比,黄原胶组兔造模膝关节滑液中IL-1β、TNF-α、PG E2和NO的含量均下降(P<0.01);黄原胶组和玻璃酸钠组中TNF-α、PG E2和NO的含量没有显著性差异(P>0.05),但黄原胶组的IL-1β含量略高于玻璃酸钠组的含量(P<0.05)。结论关节腔注射黄原胶有可能成为治疗骨关节炎的新方法。
Objective To investigate the effects of intra-articular(IA) injection of xanthan gum(XG) on the contents of interleukin-1β(IL-1β),tumor necrosis factor-α(TNF-α),prostaglandin E2(PG E2) and nitric oxide(NO) in synovial fluid in rabbits with knee osteoarthristis(OA).Methods The rabbit knee OA model was induced through IA injection of papain.Then the rabbits were randomly divided into three groups including IA injections of NaCl,sodium hyaluronate(SH) and XG.After treatment,the contents of IL-1β,TNF-α and PG E2 in synovial fluid were measured by enzyme-linked immunosorbent assay,the content of NO was measured by nitrate reductase assay.Results The contents of IL-1β,TNF-α,PG E2 and NO in synovial fluid in XG group were significantly lower than those in saline group(P0.01).The content of IL-1β in XG group was slightly higher than that in SH-treated group(P0.05),but the contents of TNF-α,PG E2 and NO were no significant differences between the two groups(P0.05).Conclusion IA injection of XG can be probably a new effective therapeutic method for OA.
