摘要
目的:探索人工蛹虫草培养基残基中虫草素的纯化技术,为进一步工业纯化打下基础。方法:经收割子实体的人工蛹虫草培养基残基先用石油醚脱脂,残渣干燥后以热乙醇提取得浸膏,浸膏转水溶后调节等电点,再用活性炭脱色,脱色后的溶液最后过732阳离子交换树脂以NH_3·H_2O洗脱液洗脱。结果:所得虫草素纯度约为80%,总得率约为1‰。结论:该提取方案设计合理、操作简便,便于工业化生产。
Objective: To study the isolation of Cordycepin from Cordyceps militaris culture medium for the further industrial process. Method: The extraction process included four steps, namely, degrease, isoelectric point regulation, absorbing decoloration and column chromatography. Firstly the culture medium of Cordyceps militaris was degreased by petroleum ether, then the residue was extracted by ethanol to afford crude fraction. The crude fraction dissolved in distilled water and then treated with isoelectrie point regulation by HCI, absorbing decoloration by active carbon. Finally the liquid was dealt with 732 cation exchange resin eluted by NH3H20, Result. Cordyeepin was obtained by these four steps above, and the purity of eordyeepin was about 80%, the total extraction rate was about 1‰. Conclusion: Tile extraction process is well designed and is easy to operate for its industrial exploitation.
